Charlo made his professional debut on August 12, 2008, against Cimmaron Davis and won by technical knockout. Thereafter, he faced Deon Nash and won by unanimous decision. He also fought against Puerto Rican Carlos Garcia. Over the next five years, he remained undefeated. 

On March 28, 2015, he went up against Michael Kenneth Finney and won by unanimous decision. Thereafter, he won against Cornelius Bundrage to capture the IBF light middleweight title.

He went up against Wilky Campfort for his title defence winning by a technical knockout. Charlo also fought against Austin Trout winning by a 12-round unanimous decision. His next title defence came against Julian Williams eating him by a technical knockout.
